Composition

Properties

Melatonin is a substance that is mainly synthesized and secreted by the pineal gland from the amino acid tryptophan in humans at night, it plays the role of a biological clock for our body and regulates immuno-endocrine and other functions of the body. Also, the ability to synthesize melatonin has been found in organelles of many tissues, including the intestinal mucosa, brain, retina, lens, trachea, skin, liver, kidney, thyroid gland, pancreas, thymus, spleen and reproductive tissues. At the same time, melatonin is an antioxidant in the skin that prevents the harmful effects of ultraviolet radiation and photodamage to the skin and is protective in conditions of chronic skin inflammation, such as atopic dermatitis.

Melatonin secretion is stimulated by the reduction in light intensity and the onset of darkness. Normal melatonin secretion is suppressed even by relatively dim light when the pupils are dilated. This is thought to be the main reason why prolonged use of devices such as laptops and smartphones before bedtime can have a negative impact on melatonin secretion, circadian rhythms and sleep. Normally, the peak of secretion is observed between 2 and 4 am, after which it gradually returns to daytime levels by the time we wake up. Melatonin secretion levels gradually increase from childhood to adolescence and show a plateau during puberty, and then slowly decrease with age, starting in the late second decade of life, reaching minimum levels after 50 years. In addition to aging, melatonin secretion can be affected by diseases, diet, environmental factors such as bright light at night, medication, and lifestyle (see Figure 1 “Age-related changes in melatonin secretion in humans”).

Circadian rhythms, which include melatonin secretion, contribute to maintaining overall health and the normal function of the body's immune, reproductive, and nervous systems.

In addition to its well-known role in sleep regulation, melatonin exhibits antioxidant activity - it protects DNA, proteins and lipids from the destructive effects of oxidative stress. The mechanism of antioxidant action of melatonin lies in its ability to neutralize free radicals that appear as a result of lipid peroxidation, as well as in its ability to activate glutathione peroxidase - a powerful endogenous factor of protection against oxidation by free radicals. Its unique feature is a cascade reaction with free radicals and the ability to respond under conditions of moderate oxidative stress. Studies have shown that melatonin surpasses in its ability to “quench” reactive oxygen species such as well-known antioxidants as glutathione, mannitol and vitamin E. In addition, unlike most intracellular antioxidants, which are localized mainly in individual cell structures, the presence of melatonin and its antioxidant activity are found in all cell structures, including the nucleus.

The antioxidant effect explains the anti-aging properties of melatonin - neutralization of free radicals prevents DNA damage and cell death. Melatonin is able to slow down age-related changes in the ovaries and maintain normal reproductive function through several mechanisms (support of telomeres, stimulation of the expression of sirtuins (SIRT, regulatory proteins with specific intracellular localization and biological functions associated with mitochondrial energy homeostasis, antioxidant activity, proliferation and DNA repair) and ribosome functions), improve well-being in women in the climacteric period (according to researchers, during menopause, 25 to 50% of women experience some sleep-related problems compared to 15% in fertile age).

Melatonin’s ability to cross cell membranes and reach cellular organelles such as mitochondria is critical for maintaining mitochondrial homeostasis. Loss of mitochondrial function, the key organelles responsible for cellular energy production, can lead to excessive fatigue and other symptoms that are common complaints in almost every chronic disease. Melatonin protects mitochondria by scavenging reactive oxygen species, inhibiting mitochondrial membrane permeability, and activating uncoupling proteins. Thus, melatonin maintains optimal mitochondrial membrane potential and preserves mitochondrial function. In addition, mitochondrial biogenesis and dynamics are also regulated by melatonin.

Finally, melatonin is involved in many activities, including genomic regulation, modulation of inflammatory and immune cytokines, directly influencing both systemic and acute anti-inflammatory responses.

Melatonin has shown indirect effects on the cardiovascular system through its regulation of heart rate and its ability to reduce nocturnal blood pressure in patients with hypertension. In addition, melatonin may serve as a potent protective agent in the cardiovascular system, due to its ability to “quench” free radicals, reduce oxidative stress, modulate metabolic activity, regulate cytokine production, and prevent apoptosis. Patients with hypertensive disorders are often prescribed beta-blockers, which can block endogenous melatonin secretion. Consequently, patients with hypertension may experience insomnia as a side effect of beta-blockers. The use of exogenous melatonin supplements may improve the quality and duration of sleep in these patients.

Indication

The dietary supplement MELATONIN-A FORTE can be used in dietary diets recommended by a doctor as an additional source of melatonin.

Melatonin contributes to:

reducing the time it takes to fall asleep;

alleviation of specific sensations when changing time zones;

falling asleep quickly and having a healthy sleep;

normal functioning of the immune system;

normal functioning of the antioxidant tissue protection system.

Recommendations for use

Adults: 1 tablet per day 30 minutes (min) before bedtime, washed down with plenty of water. Other amounts should be agreed with a doctor. Before use, a doctor's consultation is required.

Duration of use: 1-2 weeks. In the future, the possibility of repeating the course of use should be agreed with the doctor.

Do not exceed the recommended daily intake (serving). Not recommended for people with hypersensitivity to any component of the product. Given that the dietary supplement causes drowsiness, you should refrain from driving vehicles and performing other tasks that require concentration for 5 hours (h) after consuming the product.
